Molecular Cloning Workshop

This two-day molecular cloning workshop offers a brief yet comprehensive introduction to essential cloning techniques. Participants will engage in hands-on experience with key cloning procedures, learning the principles of DNA manipulation and cloning. By the workshop’s conclusion, attendees should be equipped to carry out basic molecular cloning experiments.

Day-1: Understanding the Basics

  • 9:30 AM – 11:00 AM: Welcome address – Molecular cloning techniques: key concepts and workflow
  • 11:00 AM – 12:30 PM: Hands-on training: In silico design of your cloning experiment (computer lab) – Identifying gene of interest and vectors – Selection of restriction enzymes – Constructing the primers design • In class interactive virtual cloning quiz
  • 12:30 PM – 1:00 PM: Break
  • 1:00 PM – 4:30 PM: Hands-on training: vector and DNA insert preparation and quality control •-

Day-2: Hands-on restriction/ligation cloning

  • 10:00 AM – 12:30 PM: Hands-on training: technical tips and tricks for vector selection and preparation for restriction/ligation cloning: – Intro to restriction/ligation cloning protocol – Hands-on experience with restriction enzyme digestion – Analyzing restriction digests by gel electrophoresis – Purification of digested vector for ligation – Ligating the insert into a plasmid vector
  • 12:30 PM – 1:00 PM: Break
  • 1:00 PM – 2:30 PM: Introducing recombinant plasmids into a recipient host and selection of transformants Bacterial Transformation: Transforming competent E. coli with the ligated plasmid – Plating transformed cells on agar plates – Methods of selection of transformants

  • 2:30 PM – 4.30 PM: Applications of molecular cloning : Case study 1: Identification of bacterial hypothetical systems. 

  • Theoretical Session 3  (1.5 hrs.) To be arranged: Case study 2: Construction of antibody display libraries.
